Antioksidan ve antiproliferatif etkileri bilinen isoflavonlar, potansiyel antitümörajanlar olarak görülmektedir, ancak moleküler etki mekanizmaları henüz tam olarakanlasılamamıstır. Son çalısmalar, isoflavon genistein'in, degisik hücresel olaylardagörevli birçok genin ekspresyonu üzerinde düzenleyici etkisi oldugunu göstermistir.nsülin-benzeri büyüme faktörü reseptörü 1 (IGF-1R), transmembran bir tirozinkinazdır ve proliferatif, antiapoptotik-etkili IGF sisteminde rol alır. Ligandı (IGF-I veII) ile uyarılmasını takiben aktif hale gelen IGF-1R, hücre içindeki belli proteinlerifosforlayarak, hücre proliferasyonunu uyarıcı ve apoptozu engelleyici belirli sinyalyollarını uyarır. IGF-1R düzeyinin normal hücrelerde degisik hücresel faktörlercedüzenli bir sekilde kontrol edilmesine ragmen, tümör hücrelerinde yükseldigibilinmektedir. IGF-1R ekspresyonunun genistein-etkili inhibisyonunun prostat kanserhücre hatlarında gösterilmesi, belki de, isoflavonların kanser hücreleri üzerindeki yenibir etki mekanizmasını olusturmaktadır. Bu çalısmada, genistein'in MCF-7 ve T47-Dmeme kanseri hücre proliferasyonuna doz- ve zaman-bagımlı etkisi tripan mavisi veMTS testleri ile analiz edilmis ve çesitli sinyal ileti yollarının bu etkide rolleriarastırılmıstır. Düsük genistein konsantrasyonlarının, ilk 48 saatlik evrede hücreproliferasyonunu uyardıgı, buna karsın yüksek konsantrasyonlarda zaman-bagımlınegatif etkisinin oldugu gözlenmistir. PI3K ve MAPK sinyal ileti yollarınıninhibisyonunun, düsük konsantrasyonlarda genistein-uyarılı hücre proliferasyonunumodüle ettigi gözlenmistir. Ayrıca, genistein'in IGF-1R'nün ekspresyonuna potansiyelengelleyici etkisi ve bu etkide belirli sinyal yollarının rolleri, MCF-7 hücrelerinde,RT-PCR yöntemiyle incelenmistir. IGF-1R mRNA düzeyinin genistein uygulamasıylaazaldıgı ve bu negatif etkinin, MAPK inhibitörü PD98059 varlıgında bastırıldıgıgözlenmistir.

Isoflavones, known to possess antioxidant and antiproliferative properties, havebeen shown to become potential antitumor agents. However, the mode of action ofthese compounds has not been clearly understood. Recent studies have shown thatgenistein, an isoflavone, had regulatory effects on the expression of a number of genesinvolved in various cellular events. Insulin-like growth factor-1 receptor (IGF-1R) is amembrane-bound tyrosine kinase and part of IGF system that was shown to haveproliferative and antiapoptotic activities. Upon activation by its ligand (IGF-I ve II),IGF-1R phosphorylates certain proteins within a cell, thereby inducing signallingcascades resulting in cell proliferation and the inhibition of apoptosis. Although IGFIRlevel is tightly regulated by the concerted action of various cellular factors innormal cells, it is known to increase in cancerous cells. Genistein-induceddownregulation of IGF-1R expression has been shown in prostate cancer. This may bea new aspect of the mechanism of action of isoflavones on tumor cells. In this study,the dose- and time-dependent effect of genistein on the proliferation of the breastcancer cells MCF-7 and T47-D was analyzed by tripan blue and MTS assays. It wasobserved that, at the first 48 hour period, low genistein concentrations incuced cellproliferation, whereas at high concentrations, proliferation was negatively affected in atime-dependent manner. The modulation of the low genistein-induced cellproliferation by the inhibition of the PI3K and MAPK signalling pathways was alsoobserved. The potential inhibitory effect of genistein on IGF-1R expression and therole of various signalling pathways was investigated in MCF-7 cells using RT-PCRmethod. It was oberved that the IGF-1R mRNA levels were reduced by genistein andthis negative effect was repressed in the presence of PD98059, a MAPK inhibitor. |
