Günümüzde yaygın olarak kullanılmakta olan petrol türevli plastikler,
hammaddelerinin sınırlı, yenilenemez olması ve doğada uzun süreler bozunmadan
kalabilmeleri gibi kanıtlanmış pek çok çevresel sorun ve dezavantajlarına rağmen
kolay şekil alma, elastikiyet gibi fiziksel özellikleri sebebiyle bugün halen en yaygın
kullanıma sahip plastik türüdür. Petrol bazlı plastikler yüksek dayanıklılık ve düşük
maliyete sahip olmalarına rağmen doğal ekosistemlere olan olumsuz etkileri, petrol
türevli plastiklere alternatif sağlayabilecek, plastiklerden beklenen fiziksel ve
kimyasal özellikleri taşıyan, ekolojik yönden yararlı, karbon ayakizi düşük,
biyobozunur materyallerin geliştirilmesini ve yeni kaynak arayışlarını gündeme
getirmiştir. Biyo-bazlı polimerler ve bunlardan elde edilen biyoplastikler, petrol
kökenli plastiklerin alternatifleri olarak değerlendirilmektedir. Dünya genelinde
yaklaşık 325 milyon ton fosil kaynaklı plastik üretilirken, yüksek maliyetleri
nedeniyle biyoplastikler yıllık toplam plastik üretiminin sadece yüzde 1,5‟ini
oluşturmaktadır.
Bu sebeple bu yüksek lisans tez projesinin amacı biyoplastiklerin dünyada halen
yeterli endüstriyel üretim seviyesine ulaşamama sebebi olan yüksek üretim
maliyetini düĢürmeye yönelik alternatifler geliştirerek, atıkların faydalı ürüne
dönüştürülmesidir. Bu amaca yönelik olarak biyoplastiklerin hammaddesi olan
polihidroksialkanoat (PHA) üretiminde, evsel ve endüstriyel nitelikte atıksulara
aklime olmuş aktif çamur sistemlerinden elde edilen PHA depolayan
mikroorganizmalar kullanılarak, yüksek maliyet kalemlerinden biri olan karbon
kaynağının atıksudan sağlanması ile PHA üretiminde optimum koşulların
belirlenmesi amaçlanmıştır. Çalışma 2 aşamada yürütülmüştür.
Çalışmanın ilk aşamasında evsel nitelikteki sentetik atıksu ile beslenen, çamur
bekletme süreleri değişiklik gösteren (5, 10 ve 20 gün) laboratuvar ölçekli kontrol
reaktörlerinden ve endüstriyel atıksu ile beslenen (ƟX= 6 gün) laboratuvar ölçekli
kontrol reaktöründen alınan çamurlar ile kesikli PHA depolama deneyleri ve
respirometrik analizler gerçekleştirilmiştir.
Deneysel çalışmanın ikinci bölümü ise Marmara Bölgesi‟nde yer alan 4 farklı ileri
biyolojik atıksu arıtma tesislerden alınan çamurlar ile depolama mekanizmalarının
belirlenmesi için respirometrik deneyler gerçekleştirilmiştir.
PHA üretiminde en yüksek verim karbon kaynağı olarak asetatın kullanıldığı sentetik
atıksuya aklime olmuş biyokütlenin 0,78 mg PHB(KOİ)/mg KOİ içeriği olmuştur.
Evsel nitelikteki sentetik atıksu (pepton karışımı) ile beslenen reaktörlerden alınan
çamur bekletme süreleri 5, 10, ve 20 gün olan çamurlarda depolamanın olmadığı
görülmüştür. Yüksek KOİ içeriğine sahip endüstriyel atıksuya aklime olmuş kontrol
reaktöründen alınan çamurlara asetik asit, laktik asit, propiyonik asit ve endüstriyel
atıksuyu eklenerek yürütülen kesikli deneylerde ise sırasıyla 0,70; 0,49; 0,60 ve 0,52
mg PHB(KOİ)/mg KOİ‟lik depolama verimi hesaplanmıştır. Dört farklı atıksu arıtma
tesisinden alınan çamurlarla yürütülen kesikli deneylerde ise depolama
görülmemiştir.
Bu çalışmada, organik asit içeriği yüksek atıksuların hammadde kaynağı olarak
kullanılmasıyla, PHA depolama kabiliyeti olan aktif çamurun bulunduğu atıksu
arıtma tesislerinde biyopolimer üretimi yapılabileceği gösterilmiştir.

Petroleum-derived plastics are still the most widely used plastic types due to their
physical properties such as easy forming and elasticity despite their proven
environmental problems and disadvantages such as their raw materials being limited,
non-renewable and long-term degradation in nature. Although petroleum-based
plastics have high durability and low cost, due to their negative effects on natural
ecosystems, biodegradable materials that can provide alternative to petroleum-
derived plastics, which have the expected physical and chemical properties of
plastics, have been brought to the agenda for new resources. Bioplastics derived from
bio-based polymers are considered as alternatives to petroleum-based plastics. While
around 325 million tons of fossil-based plastics are produced worldwide, bioplastics
account for only 1.5 percent of total annual plastic production due to their high cost.
Biopolymers used in the production of bioplastics can be synthesized by chemical
methods or produced biotechnologically from microorganisms or plants. In the
production of commercial PHA, expensive raw materials and chemicals are used as
organic material sources. Many laboratory-scale study alternatives have been
proposed to increase the profitability of the system and facilitate its application in the
plastic market. One of the most promising alternatives is the use of industrial by-
products or waste water as a carbon source for PHA production. Using waste as a
carbon source, PHA is considered as an environmentally friendly management
method. Since raw material is the main carbon source for PHA synthesis and
bacterial growth, the use of organic wastes as a source of organic material is a
promising alternative to a reduction in PHA production costs. In the literature, it is
proposed to integrate PHA production with biological wastewater treatment and also
provide benefits such as resource recovery and sludge minimization. Today,
activated sludge systems are a widely used technology in biological wastewater
treatment. The main approach in activated sludge systems is to remove contaminants
from wastewater by biochemical reactions as in other biological treatment systems.
In biological systems, the removal of these pollutants originating from wastewaters is
carried out by microorganisms that take up pollutants to produce cell main
components and use the resulting energy. In the absence of the organic carbon
required for growth and energy, the internal respiration phase is also carried out,
where many biochemical activities are performed. In addition to the removal of
organic matter in the design and modeling of activated sludge systems, the storage of
recently developed organic carbon as a polymer in the cell is considered as an
important process.
For this reason, the aim of this master thesis project is to convert wastes into useful
products by developing alternatives to reduce the high production cost, which is the
reason why bioplastics still do not reach a sufficient level of industrial production in
the world. For this purpose, in the production of polyhydroxyalkanoate (PHA),
which is the raw material of bioplastics, it is aimed to determine optimum conditions
in PHA production by providing carbon source, which is one of the high cost items,
by using microorganisms which are obtained from activated sludge systems obtained
from domestic and industrial wastewaters. The study was conducted in 2 stages.
In the first stage of the study, batch PHA storage experiments and respirometric
analyzes were performed with sludges taken from laboratory scale control reactors
fed with domestic synthetic wastewater and whose sludge retention times varied (5,
10 and 20 days) and laboratory scale control reactor fed with industrial wastewater.
In the second part of the experimental study, respirometric experiments were carried
out to determine the sludges and storage mechanisms taken from 4 different
advanced biological wastewater treatment plants in the Marmara Region.
The highest yield in PHA production was 0.78 mg PHA(COD)/mg COD content of
biomass which was acclimate to synthetic wastewater using acetate as carbon source.
It was observed that there was no PHA storage in sludges with sludge retention times
of 5, 10, and 20 days taken from reactors fed with domestic synthetic wastewater
(peptone mixture). In batch experiments carried out by adding acetic acid, lactic acid,
propionic acid and industrial wastewater to the sludges taken from the control reactor
which has been acclimated in industrial wastewater with high COD content, storage
efficiency of 0.70, 0.49, 0.60 and 0.52 mg PHA(COD)/mg COD were calculated
respectively.
In this study, it has been shown that biopolymer production can be realized in
wastewater treatment plants with active sludge having PHA storage capacity by
using wastewater (especially high acetate content) as raw material source. |
