Bu çalışmada, 3-Fenil-5-sülfamoil-N-(7/8/9-nonsübstitüe/sübstitüe-3-okso-1-tiya-4-azaspiro[4.4]non/[4.5]dek-4-il)-1H-indol-2-karboksamid (4-12) yapısında yeni bileşikler sentezlenmiş ve bileşiklerin antimikrobiyal aktivitesi ve NRK-52E sıçan böbrek epitel hücrelerinde sitotoksisitesi araştırılmıştır. Bu amaçla sülfanilamid' in HCl' li ortamda sodyum nitrit ile reaksiyonundan oluşan 4-sülfamoilbenzendiazonyum klorür' ün etil 2-benzilasetoasetat ile etkileştirilmesinden etil 2-benzil-2-(4-sülfamoilfenil)hidrazono]asetat (1) elde edilmiş; 1'in Fischer-indol yöntemine göre HCl'li ortamda ısıtılmasıyla etil 3-fenil-5-sülfamoil-1H-indol-2-karboksilat (2) kazanılmıştır. 2' nin hidrazin hidrat ile reaksiyonundan 3-fenil-5-sülfamoil-1H-indol-2-karbohidrazid (3) oluşturulmuş. Spirotiyazolidinon türevi 3-fenil-5-sülfamoil-N-(7/8/9-nonsübstitüe/sübstitüe-3-okso-1-tiya-4-azaspiro[4.4]non/[4.5]dek-4-il)-1H-indol-2-karboksamid (4-12) numaralı bileşikler 3' ün uygun siklik ketonlar ve merkaptoasetik asid ile siklizasyonu sonucu elde edilmiştir. Bileşiklerin oluşum mekanizmaları tartışılmış ve yapıları elementel analiz, UV, IR, 1H-NMR, HSQC ve ESI-MS verileri ile doğrulanmıştır. Bileşiklerin (4-12) antimikrobiyal aktiviteleri Staphylococcus aureus ATCC 6538, Staphylococcus epidermidis ATCC 12228, Escherichia coli ATCC 25922, Klebsiella pneumoniae ATCC 4352, Pseudomonas aeruginosa ATCC 27853, Proteus mirabilis ATCC 14153 ve Candida albicans ATCC 10231'e karşı test edilmiştir. Bileşiklerin Staphylococcus epidermidis ATCC 12228 ve Pseudomonas aeruginosa ATCC 27853 karşı zayıf etki gösterdikleri saptanmıştır. Tüm bileşiklerin NRK-52E sıçan böbrek epitel hücrelerinde MTT testi ile sitotoksisitesi araştırılmıştır ve çalışılabilen maksimum konsantrasyonda sitotoksik etki saptanmamıştır. This study describes the synthesis of novel 3-phenyl-5-sulfamoyl-N-(7/8/9-nonsubstituted/substituted-3-oxo-1-thia-4-azaspiro[4.4]non/[4.5]dec-4-yl)-1H-indole-2-carboxamide (4-12) derivatives and their subsequent testing for both antimicrobial activity and cytotoxcicity against rat kidney epithelial cells (NRK-52E). In the course of the syntheses 4-sulfamoylbenzenediazonium chloride, obtained from the reaction of sülfanilamide with sodium nitrite and HCl, was reacted with ethyl 2-benzylacetoacetate to afford ethyl 2-benzyl-2-(4-sulfamoylphenyl)hydrazono]acetate (1). 1 was cyclized to ethyl 3-phenyl-5-sulfamoyl-1H-indole-2-carboxylate (2) employing the Fischer-indole procedure. Reaction of 2 with hydrazine hydrate yielded 3-phenyl-5-sulfamoyl-1H-indole-2-carbohydrazide (3). Spirothiazolidinone derivatives 3-phenyl-5-sulfamoyl-N-(7/8/9-nonsubstituted/substituted-3-oxo-1-thia-4-azaspiro[4.4]non/[4.5]dec-4-yl)-1H-indole-2-carboxamide (4-12) were obtained by cyclization of 3 and appropriate cyclic ketones with mercaptoacetic acid. The reaction mechanisms involved in the formation of the end products have been discussed and the structures of the new compounds were confirmed by the data obtained from elemental analysis, UV, IR, 1H-NMR, HSQC and ESI-MS spectra. Compounds 4-12 have been tested for their antimicrobial activity against Staphylococcus aureus ATCC 6538, Staphylococcus epidermidis ATCC 12228, Escherichia coli ATCC 25922, Klebsiella pneumoniae ATCC 4352, Pseudomonas aeruginosa ATCC 27853, Proteus mirabilis ATCC 14153 ve Candida albicans ATCC 10231. They have been found to show weak activity against Staphylococcus epidermidis ATCC 12228 ve Pseudomonas aeruginosa ATCC 27853. All compounds have been tested with the MTT assay and showed no cytotoxicity against NRK-52E cells at the highest working concentrations.
