ÖZET "Alışılagelmişin Dışında Serolojik Profil Gözlenen Hepatit B İnfeksiyonlannda HBV DNA Araştırılması" isimli tez, hepatit B virüsünün tanısında karşılaşılan güçlükleri çözmek ve tedaviye yardımcı olmak amacıyla gerçekleştirildi. ELISA yöntemiyle hepatit B virüsü markırlan istenen hastalardan Abbott'un Makro ELISA cihazı kullanılarak HBV markırlanna bakıldı. Hepatit B markır sonuçlan arasında çelişkiler bulunan ve alışılagelmişin dışında serolojik profil gözlenen 23 'ü kadın, 27' si erkek toplam 50 hasta serumu biriktirildi. Bu 50 hasta serumu serolojik profillerine göre 9 ayn grupta toplandı. Bu serumlara Digene'nin Hybrid Capture System'i kullanılarak kantitatif olarak HBV DNA bakıldı. Değerlendirme sonunda 2 hastada HBV DNA pozitif olarak bulundu. Bu hastalardan biri HBsAg ve anti-HBs'si pozitif grup içinde, diğeride HBsAg ve HBeAg'si pozitif grup içinde yer aldı. Bu bulgular daha önce konuyla ilgili yapılan, serolojik profillerinde çelişkiler olan hastalarda HBV DNA bakılmasının önemini araştıran sonuçlan desteklemektedir. Bu çalışma ile elde edilen sonuçlar, ELISA yöntemiyle serolojik profillerinde çelişkiler gözlenen hastalardan HBV DNA bakılarak kesin tanıya ulaşılabileceğini göstermiştir. 6. SUMMARY The theme of my thesis was 'Detecting HBV DNA in hepatitis B infections with unusual serological profile'. This study was performed in order to solve some diagnostic problems in hepatitis B infection and to support treatment modalities. Requested HBV markers to be studied with the ELISA method were evaluated with the Macro ELISA apparatus of the firm Abbott. Serum samples from 23 female and 27 male, totally 50 patients with unexpected hepatitis B results and unusual profiles were collected. These serum samples from 50 patients were divided into 9 different groups according to the serological profiles. HBV DNA was studied with the Hybrid Capture System of Digene. Two patients were find to be HBV DNA positive. One patient was in the HbsAg and anti-HBs positive group; while the other patient was in the HbsAg and HbeAg positive group. These results support the studies which pointed out the importance of evaluating HBV DNA in surprising serologicol profiles. This study shows that searching for HBV DNA is helpful to make the true diagnosis in cases with surprising serological profiles by the ELISA method.
