Çocukluk çağı T hücreli akut lenfoblastik lösemisi (T-ALL) akut lenfoblastik lösemi vakalarının %10 kadarını oluşturan, patogenezi ve prognozu ile ilişkilendirilmiş sınırlı sayıda genetik değişiklik tanımlanmış malin bir T hücresi hastalığıdır.
Mikro RNA'lar (miRNA) 17-25 nükleotid uzunluğunda küçük, fonksiyonel, protein kodlamayan tek iplikli RNA'lardır. Apoptoz, farklılaşma ve proliferasyon gibi pek çok hücresel yolakta görev alırlar. MiRNA'ların birçoğu tümör oluşumuna onkogen ya da tümör baskılayıcı genleri hedef alarak etkide bulunabilirler.
Çalışma grubumuzda yapılan önceki çalışmalarda çocukluk çağı T-ALL hastalarında gen anlatımı azalan yeni bir tümör baskılayıcı gen olan, SnoN/SKIL geni tanımlanmıştır.
Bu tez çalışmasında ALL hastalığını temsil eden hücre hatlarında azalan miR-223 geni anlatımı karşısında, hedef gen konumunda bulunan SnoN/SKIL geni anlatımının %30-80 oranında arttığını tespit ettik. Bu iki gen arasındaki ilişkiyi lusiferaz çalışmalarıyla da fonksiyonel olarak doğruladık. Molt4 ve Jurkat hücrelerinde sonuçlar anlamlı veya anlamlılığa çok yakındı (p=0,04 ve p=0,06). Bunun yanısıra SnoN/SKIL geni anlatımının artmasıyla hücrelerin apoptoza geçme eğiliminin arttığını akım sitometrisi tekniği ile yaptığımız hücre canlılığı ölçümleriyle gözlemledik. Bulgularımız miR-223 ile SnoN/SKIL geni arasındaki fonksiyonel bir ilişkiyi doğrulamakta ve özellikle T-ALL grubunda miR-223'ün onkogenik, SnoN/SKIL geninin ise tümör baskılayıcı özelliklerde olduğunu göstermektedir.

Childhood T-cell acute lymphoblastic leukemia ( T- ALL) constitutes 10-15% of all ALL cases and 20-25% of adult ALL cases. Patients with T-ALL have hard phases, pathogenesis and prognosis associated with a limited number of defined mutations.
Micro-RNAs (miRNA) that are small (17-25 nucleotides), functional and non-coding RNAs which are played roles in apoptosises, cell differentations and proliferations. According to previous studies, the mutations or changing in levels of gene expressions of miRNAs are associated with many diseases, especially cancer. Also, many of the miRNAs may influence tumor formation by targeting oncogenes or tumor suppressors.
In our previous study, we have identified that the expression levels of a new tumor suppressor gene, called as SnoN/SKIL, decreases in T-ALL patients. Also, we determined that, the gene expression levels of miR-223, which targets SnoN/SKIL and is tend to effective in development of T-lymphocytes of progenitor cells to myeloid series in hematopoietic process, increases in the same patients.
In that study, we determined an increase by 30-80% in the expression of SnoN/SKIL gene in the face of miR-223 supression in cell lines representing ALL disease. Then the functional relationship between these two genes were confirmed by luciferase reporter assay. The results for Molt4 and Jurkat cell lines were significant (p=0,04 and p=0,06). We also observed that the tendency for going into apoptosis in the cell lines increased by the overexpression of SnoN/SKIL gene, via flow cytometry technique. Our results verify a functional relationship between SnoN/SKIL and miR-223. We also showed the tumor suppressor character of SnoN/SKIL gene in T-ALL while miR-223 indicates an oncogenic character. |
