Difteri hastalığı süresince kalpte miyotoksik etki oluşmaktadır. Miyotoksik etki, protein sentezinin durması ile açıklanmaktadır. Ancak protein sentezinin baskılanması ile difteri toksinine bağlı hücre ölümü arasındaki ilişki tam olarak aydınlatılamamıştır. Difteri toksininin (DT) varlığında, ökaryotik elongasyon faktörü 2'nin (eEF-2) ADP-ribozillenmesi sonucu protein sentezinin baskılandığı bilinmektedir. DT'nin protein sentezini durdurmasının yanı sıra hücre iskeletinin yıkılmasına da sebep olduğu ve hücre iskeleti yapıtaşı olan aktinin eEF-2 ile etkileştiği öne sürülmüştür. Bu çalışmada, DT alt yapıları olan, A ve B kesimleri (FA, FB) ile aktin filamentleri arasındaki olası etkileşimin araştırılması amaçlandı. Bu doğrultuda toksine duyarlı ve dirençli hücreler ile fibriler aktinin çok yoğun olduğu kalp kası dokusu kullanıldı. Değişen DT derişimleri ile farklı süreler boyunca bekletilen hücre ve dokularda, immünfloresan görüntüleme sonucu DT ile aktin filamentlerinin etkileştiği saptandı. Hücre içinde, DT ile eEF-2 dağılımının benzerliği gösterildi. DT ile bekletilen kalp kası dokusunun ince yapı düzeyinde iletim elektron mikroskobu ile görüntülenmesi sonucunda miyobriloliz, şişme, mitokondriyal hasar ve çekirdekte kromatin kümelenmesi gözlendi. Moleküler düzeyde yapılan inceleme ile DT'nin A kesimi ile aktinin kompleks proteini oluşturduğu sonucuna varıldı. Bu bulgular DT'nin, hücre iskeleti üzerinden etkili olabilecek bir sitotoksik yolağın yanında, programlanmış hücre ölümü ile ilişkili yolakları da etkinleştirebileceğini düşündürmektedir.
The myotoxicity, which occurs during the diphtheria disease, is explained by the inhibition of the protein synthesis. However the relationship between the protein synthesis inhibition and the cell death could not be fully revealed. It is known that the protein synthesis is inhibited as a result of the ADP-ribosylation of eukaryotic elongasyon factor 2 (eEF-2) in the presence of diphtheria toxin (DT). It has been put in front that DT lead also to the fall of the cell skeleton as a result of the interaction of eEF-2 and the actin. In this study, it is aimed to investigate the possible interaction between the actin filaments and the fragments (FA, FB) of DT. In this context, it is used DT sensitive and resistant cells and heart muscle tissue where the filamentous actin is abundant. The interaction of DT between actin filaments has been shown on cells and tissue by the immunefluorescent techniques with varying concentration of the toxin and incubation time. The distribution of either DT or eEF-2 has been exposed as similar. It was observed by transmission electron microscope myofibrilolysis, dilatation, mitochondrial damage and the clump of chromatin in the nucleus of myofibril under the effect of DT. The result of the molecular analysis was concluded that the fragment A and actin could form a protein complex. Those findings gave rise to thought that apoptotic pathways could become trigered besides the cytotoxic pathways that DT could act via the actin cytoskeleton.
