Bu çalısmada, 41 adet Staphylococcus aureus susu; fenotipik özellikleri, plazmit DNAve 2 farklı poliakrilamid jel elektroforez (SDS-PAGE) yöntemi kullanılarak elde edilenprotein profillerine göre karakterize edilmistir. Morfolojik, fizyolojik ve biyokimyasalözelliklerine göre, bütün susların S. aureus türüne ait oldukları belirlenmistir. Antibiyogramsonuçları test edilen susların en az iki antibiyotige dirençli oldugunu, fakat vankomisinantibiyotigine duyarlı olduklarını göstermistir.Bütün ırkların plazmit profillerini belirlemek için agaroz jel elektroforez metodukullanılmıstır. Agaroz jel elektroforez sonuçlarına göre, susların tek plazmit içerdigibelirlenmistir. SDS-PAGE tekniginin kullanıldıgı nümerik analiz sonuçları; toplam hücreprotein profilleri esas alındıgında benzerlik düzeyi %45.84'ün üzerinde 2 farklı grup ve hücredısı protein profilleri esas alındıgında ise benzerlik seviyesi %68.81'in üzerinde 4 farklı grupolustugunu ortaya koymustur. Diger taraftan, Native-PAGE tekniginin kullanıldıgı nümerikanaliz sonuçları; toplam hücre protein profilleri esas alındıgında benzerlik düzeyi %53.51'inüzerinde 3 farklı grup ve hücre dısı protein profilleri esas alındıgında ise benzerlik seviyesi%62.90'ın üstünde 2 farklı grup olustugunu ortaya koymustur. Arastırma SDS-PAGEyöntemiyle elde edilen toplam hücre ve hücre dısı protein profillerinin test edilen S. aureussuslarının taksonomik iliskilerinin incelenmesinde etkili bir yaklasım saglamadıgınıgöstermistir. Buna karsın, Native-PAGE yöntemiyle elde edilen toplam hücre ve hücre dısıprotein profillerinin ise susların ayrımında daha faydalı oldugu belirlenmistir.Anahtar Kelimeler: Karakterizasyon, PAGE, Plazmit, Protein Profilleri, S. aureus.

In the present study, 41 Staphylococcus aureus strains were characterized according tophenotypic properties, plasmid DNA and protein profiles obtained from using two differentpolyacrylamide gel electrophoresis (PAGE) methods. According to the morphological,physiological and biochemical characteristics, all strains were consisted of S. aureus species.The antibiogram results indicated that all tested strains were resisted against at least twoantibiotics, but susceptible to vancomycin.Agarose gel electrophoresis was employed to determine plasmid profiles. The agarosegel electrophoresis results confirmed that all strains consisted of only one plasmid. Thenumerical analysis of SDS-PAGE technique based on whole-cell protein patterns resultsrevealed 2 distinct groups with a similarity level of above 45.84%. The extracellular proteinpattern results revealed 4 distinct groups with a similarity level of above 68.81%. Afterwards,in the Native-PAGE technique, while whole-cell protein pattern results revealed 3 distinctgroups with a similarity level of above 53.51%, the extracellular protein pattern resultsrevealed 2 distinct groups with a similarity level of above 62.90%. The research indicated thatwhole-cell and extracellular protein profiles obtained by SDS-PAGE did not provide aneffective approach to the investigation of taxonomic relationship within the tested S. aureusstrains. On the other hand, it determined that whole-cell and extracellular protein profilesobtained by Native-PAGE were more suitable ror discrimination of the tested strains.Key words: Characterization, PAGE, Plasmid, Protein Profiles, S. aureus. |
