N-fosfonometil glisin (glifosat), kararlı karbon-fosfat (C-P) bağlı, sentetik ve seçici olmayan bir herbisit bileşiği iken 2,4-diklorofenoksi asetik asit (2,4-D), fenoksi asitler grubuna dahil olan, seçici bir herbisittir. Topraktaki 2,4-D ve glifosat kalıntılarının çevre ve insan sağlığına zararlı etkileri olduğu bilinmektedir. Bu çalışmada, Kahramanmaraş ve Balıkesir bölgelerinde, sırasıyla glifosat ve 2,4-D ile muamele edilmiş iki tarım arazisinden izole edilen bakterilerin, adı geçen herbisitleri degrade etme potansiyellerinin spektrofotometrik testler ve HPLC yardımıyla belirlenmesi hedeflenmiştir. Ayrıca, bu bakterilerin gen sekanslama ve protein analizi yöntemiyle moleküler düzeyde tanımlanması amaçlanmıştır. Bu amaç doğrultusunda; seri seçilimlerle elde edilen bakteri kolonilerinin, izole edildikleri araziye bağlı olarak karbon kaynağı olarak yalnızca üç farklı derişimde (0,1g/L, 0,5g/L ve 1g/L) glifosat ya da 2,4-D içeren besiyerinde yetiştirilmeleri sağlanmıştır. Yetiştirilen bakterilerin büyüme hızları 14. günde takip edilmiş ve bakterilerin degredasyon yeteneklerinin anlaşılması için besi ortamında kalan glifosat veya 2,4-D miktarları aynı süre sonunda ölçülmüştür. Elde edilen sonuçlara göre; izole edilen bakterilerden hiçbiri glifosatı degrede edememekte hatta bazılarında glifosat bakteri büyümesini olumsuz yönde etkilemektedir. Yine de, bu ortamda en iyi büyüme gösteren bakteriler Klebsiella variicola ve Klebsiella pneumoniae olarak tanımlanmıştır. Benzer şekilde, 2,4-D'yi degrede edebilen bir bakteri bulunamamış, ancak Acinetobacter baumannii, Acinetobacter calcoaceticus ve Stenotrophomonas sp. bakterilerinin 2,4-D'li ortamda büyüyebildikleri tespit edilmiştir.
N-phosphonomethyl glycine (glyphosate) is a synthetic and non-selective herbicide compound containing a stable carbon-phosphate (C-P) bound, while 2,4-dichlorophenoxy acetic acid (2,4-D) is a selective herbicide which is a part of the phenoxy acids group. 2,4-D and glyphosate residues in soil are known to have harmful effects on environment and human health. In this study, it was aimed to determine the degradation potential of the mentioned herbicides by the bacteria isolated from two agricultural fields in Kahramanmaraş and Balıkesir regions which were waxed with glyphosate and 2,4-D, respectively, through spectrophotometric tests and HPLC. It was also aimed to identify these bacteria at the molecular level by gene sequencing and protein analysis. In order to achieve these goals; bacterial colonies obtained by serial selection were grown in the media containing three different concentrations (0.1g/L, 0.5 g/L and 1 g/L) of glyphosate or 2,4-D as the only carbon source, depending on the land from which they were isolated. Growth rates of the bacteria were monitored on the 14th day, and the amount of glyphosate or 2,4-D remaining in the medium was measured at the same time in order to understand the degradation ability of the bacteria. According to the results obtained; none of the isolated bacteria degrades glyphosate, and in some, it adversely affects bacterial growth. Yet, Klebsiella variicola and Klebsiella pneumoniae were identified as the best growing bacteria in this medium. Similarly, no bacteria capable of degrading 2,4-D were found, however, it was observed that Acinetobacter baumannii, Acinetobacter calcoaceticus and Stenotrophomonas sp. bacteria were able to grow in the medium containg 2,4-D.
