Bu çalışmada, Ammi visnaga ve Ammi majus'un çeşitli ekstraktlarının antioksidan, antifungal ve sitotoksik aktivitelerinin belirlenmesi ve RAPD-PCR yöntemiyle filogenetik olarak karşılaştırılması hedeflenmiştir. Bitkilerin metanol ekstrelerinin antioksidan aktiviteleri beta-karoten-linoleik asit yöntemi, DPPH radikal giderme aktivitesi ve metal bağlama yöntemleri ile belirlenmiştir. Ayrıca bu ekstrelerin toplam fenolik ve toplam flavonoit içerikleri de belirlenmiştir. Bitkilerin antioksidan aktiviteleri karşılaştırıldığında A. majus'un toplam fenolik ve flavonoit içeriklerinin A. visnaga'ya göre daha yüksek olduğu saptanmasına rağmen ß-karoten linoleik asit yönteminde, A. visnaga'nın antioksidan aktivitesinin A. majus'a göre daha yüksek olduğu belirlenmiştir. DPPH yöntemi sonuçları her iki bitkinin de antioksidan aktivitelerinin yakın olduğunu gösterirken, A. majus'un metal bağlama etkisininA. visnaga'ya göre daha yüksek olduğu gözlenmiştir. A. majus ve A. visnaga'nın metanol, hekzan ve kloroform ekstrelerinin Aspergillus niger üzerine antifungal etkileri incelenmiştir ve bu ekstreler, A. niger'in gelişimini engelleyememiştir. A. visnaga (kök) metanol ekstresi ve A. visnaga (tohum) metanol ekstresi, tümör hücrelerin %50 ve/veya üstü oranında, khellin ise DU-145 ve PC-3 hücreleri hariç diğer hücrelerin %50 ve/veya üstü oranında gelişimini engellemiştir. A. majus'tan elde edilen uçucu yağ, sadece MCF-7 hücresinde etki göstererek, hücrelerin %88'inin gelişimini engellemiştir. A. majus (kök) metanol ekstresi test edilen 4 hücre tipinde de etki göstermemiştir. Ancak A. visnaga'nın A. majus'a göre fenolik ve flavonoit içeriklerinin düşük olduğu görülmektedir. Bu sonuç, sitotoksik aktivite'den fenolik ve flavonoit bileşiklerin sorumlu olmadığını göstermektedir. Bitkiler moleküler düzeyde RAPD yöntemi ile karşılaştırmalı olarak incelenmiştir. A. visnaga (Selam) ve A. majus arasındaki polimorfizm değeri % 0.041, A. visnaga (Selam) ve A. majus arasındaki polimorfizm değeri % 0.178, A. visnaga (Belen) ve A. majus arasındaki polimorfizm değeri ise % 0.65 bulundu. Buna göre A. visnaga (Belen) ve A. visnaga (Selam) örnekleri arasında %91, A. majus ve A. visnaga (Selam) örnekleri arasında %89, A. majus ve A. visnaga (Belen) arasında %81 benzerlik bulunmaktadır. Çalışmada kullanılan 20 primerin, örneklerde oluşturduğu polimorfik bantlar dikkate alınarak Sorensen indeksine göre filogenetik ilişkinin belirlendiği dendrogram hazırlanmıştır. 20 primerden sadece 4 primerin değerlendirilmesiyle ortaya çıkan bu sonuçların, polimorfik bantlar meydana getirecek daha fazla sayıda primer ile tekrarlanması gerekmektedir. Elde edilen verilerin, ileride yapılacak çalışmalara temel oluşturacağı düşünülmektedir.
In this study, it is targeted that determination of antioxidant, antifungal and cytotoxic activities of Ammi majus and Ammi visnaga various extracts and phylogenetic relationship of the species by RAPD-PCR method. Methanol extracts of A. visnaga andA. majus were investigated for their antioxidant capacity by three different assays, namely, the ß-carotene linoleic acid systems, DPPH radical scavenging and metal chelating activities. In addition, total phenolics and total flavonoid contents of these methanol extracts were determined. Total phenolics and flavonoid content of A. majus was found to be higher than A. visnaga. However, antioxidant activity of A. visnaga was higher than A. majus in ß-carotene linoleic acid systems. The DPPH test system results of A. majus and A. visnaga was found to be similar. Metal chelating potential of A. majus is higher than A. visnaga. Methanol, hexane and chloroform extracts of A. majus and A. visnaga were assayed for antifungal activity against Aspergillus niger and these extracts did not inhibit A. niger. Methanol extract of A. visnaga (root) and A. visnaga (seed) inhibited 50% or over of tumor cells, khellin inhibited other cells 50% or over of tumor cells except DU- 145 and PC-3 cells. Essential oil of A. majus affected only on MCF-7 cell by inhibiting 88% of tumor cells while A. majus (root) methanol extract did not inhibit four tested cells type. But A. visnaga?s phenolic and flavonoid contents are found to be lower in comparison with to those of A. majus. According to this result, phenolic and flovonoid compounds are not responsible for the cytotoxic activity. Plants were comparatively investigated at the molecular level by RAPD technique. Between A. visnaga (Selam) and A. visnaga (Belen) polymorphism value was determined to be 0.041%, between A. visnaga (Selam) and A. majus polymorphism value was determined to be 0.178%, between A. visnaga (Belen) and A. majus polymorphism value was determined to be 0.65%. According to these results, the similarity rate was found as 91% between the samples of A. visnaga (Belen) and A. visnaga (Selam); 89% between the samples of A. majus and A. visnaga (Selam); while it was found as 81% between the samples of A. majus and A. visnaga (Belen). In this study, a dendrogram which shows pylogenetic relationship based on Sorensen index, was prepared regarding to the polymorphic bands, formed by used 20 primers on the samples. These results consisting of 4 primers? evaluation out of 20 primers, are considered to be repeated with more primers which will form polymorphic bands. Data obtained here are expected to constitute a base for the further studies.
