Bu çalışmanın amacı, besinlerde antimikrobiyal madde olarak kullanılanBifenil'in genotoksik etkisinin olup olmadığını insan periferal lenfositlerinde kardeşkromatid değişimi (KKD), kromozom anormalliği (KA) ve mikronükleus (MN)testleri ile araştırmaktır. Bu amaçla insan periferal lenfositleri bifenil'in 4 farklıkonsantrasyonu (10, 30, 50 ve 70 ?g/ml) ile 24 ve 48 saat boyunca muameleedilmişlerdir.Bu çalışmada farklı konsantrasyonlardaki bifenil ile 24 ve 48 saatlik sürelerdemuamele edilen insan periferal lenfositlerinde KKD, KA ve MN sayısı hemmuamelesiz kontrole hem de çözücü kontrole (DMSO) göre istatistiksel olarakönemli derecede artmıştır. Bu artışın genellikle doza bağlı olduğu görülmüştür.Ancak bu artışlar pozitif kontrol olan MMC'deki kadar olmamıştır. Bifenil, sadecekromozom yapı anormalliğini artırmış, kromozom sayı anormalliğini iseartırmamıştır. Bu çalışmada bifenil, hem 24 saatlik hem de 48 saatlik muamelesürelerinde replikasyon indeksi (RI) ve nükleer bölünme indeksi (NBI)'ni her ikikontroldekine göre önemli ölçüde düşürmüş ve bu düşüş doza bağlı olarakgerçekleşmiştir. Fakat bifenil mitotik indeks (MI)'i etkilememiştir.

The aim of this study was to determine the possible genotoxic effects ofbiphenyl, which is used as an antimicrobial agent in food, by using sister chromatidexchanges (SCEs), chromosome aberrations (CAs) and micronucleus (MN) tests inhuman peripheral lymphocytes. The human peripheral lymphocytes were treatedwith four concentrations of biphenyl (10, 30, 50 ve 70 ?g/ml) for 24 and 48 hourstreatment periods.In the present study, biphenyl significantly increased the frequency of SCEs,CAs and the frequency of MN when compared with both untreated control andsolvent (DMSO) control. The induction of these abnormalities were in a dosedependentmanner. However, the potency of biphenyl on induction of SCEs, CAsand MN were lower than that caused by the positive control MMC. Biphenyl wascapable to induce the structural CAs instead of numerical CAs. Biphenyl alsoshowed a cytotoxic effect by decreasing the replication index (RI) and nucleardivision index (NDI) in a dose dependent manner for both 24 and 48 hours treatmentperiods. However, biphenyl did not affect the mitotic index (MI). |
