Kurşun, çinko gibi geçmişten beri yaygın kullanıma sahip olan ağır metallerin kullanımları sırasında çevreye ve dolayısıyla insan sağlığına olumsuz etkilerinden dolayı ağır metallerin önemi giderek artmaktadır. Kurşun doğaya geniş ölçüde yayılmıştır. Yakıtlarda ve boya pigmenti gibi çok yaygın olarak kullanılması insanlarda sağlık ve çevre problemlerine neden olmuştur. Kurşun vücutta biriken ciddi bir zehirdir. Vücudumuza hava, su ve yiyeceklerle girer. Kurşun enzim inhibitörü olarak ?SH gruplarına bağlanmaktadır. Kurşun kalsiyum metobolizması gibi kemiklerde depolanmaya başlamaktadır. Tetrametil kurşun gibi organik kurşun bileşikleri, deriden hızlıca absorbe edildiklerinden dolayı oldukça zehirlidirler. İnsan akut kurşun zehirlenmelerinde böbreklerde, karaciğerde, beyinde, üreme ve merkezi sinir sistemin de yüksek hasara bazende ölüme neden olmaktadır. Hafif kurşun zehirlenmeleri ise kansızlık ve baş ağrısına neden olmaktadır. Tüm bu bulgular sebebiyle az miktardaki metal iyonlarının hassas bir şekilde tayin edilmesi ilgi çekmektedir. Bu maddeler üzerine yapılmış çalışmalar genellikle çeşitli ligandlar ile yaptıkları komplekslerin yapı aydınlatmaları üzerinedir. En yaygın kullanılan ağır metallerden kurşun için spektrofotometrik tayin yöntemleri oldukça azdır. Bu bağlamda bu tez çalışmasının amacı; Kurşun için hızlı ve kolay uygulanabilir spektrofotometrik yöntemlerin geliştirilmesidir. Kurşun için geliştirilen spektrofotometrik yöntem kurşun iyonlarının 1,5-Bis (salisiliden)tiyokarbohidrazon (BSTH) ile kompleks oluşturması ve oluşan kompleksin 419 nm'de spektrofotometrik tayinine dayanmaktadır. Geliştirilen yöntemlerin analitler için molar absorptivite ve LOD (belirtme sınırı) ile LOQ (tayin sınırı) gibi analitik performans özellikleri şöyledir: Kurşun için ?=1,20x104 L mol-1cm-1; LOD=1,8x10-6 mol L-1 ve LOQ=6x10-6 mol L-1' dir. Çinko için ise ?=2,54x104 L mol-1cm-1; LOD=2,36x10-7 mol L-1 ve LOQ 7,88x10-7 mol L-1'dir. Bu çalışmada Pb(II) tayininde girişim etkileri oluşturabilecek Ni(II), Cd(II), Zn(II), Ag(II), Cu(II), Hg(II) ve Fe(III) gibi diğer metal iyonlarının bu etkilerini kontrol altında tutup engellemek için NaCN kullanılmıştır ve optimum koşullar altında çeşitli yabancı iyonların etkileri 10-3M kurşuna karşı ayrı ayrı hesaplanmıştır.
Since the use of conventional heavy metals like lead and zinc have given rise to unforeseen accidents the negative impact to people and environment for heavy metals is on the rise. Lead is widely distributed in nature. The most widespread uses, which have led to environmental and health problems, are its use in gasoline and paint pigments. Lead is a serious cumulative body poison. It enters our body system through air, water, and food. Lead (Pb2+) binds itself with the -SH group in enzymes or proteins and acts as an enzyme inhibitor. Lead interferes with the calcium metabolism and becomes deposited in bone. Organic lead compounds, such as tetramethyl lead, are highly poisonous because they are readily absorbed by the body through skin. Acute lead poisoning in humans causes severe damage in the kidneys, liver, brain, reproductive system, and central nervous system, and sometimes causes death. Mild lead poisoning causes anemia, headache. All of these findings cause great concern regarding public health, demanding the accurate determination of this metal ion at trace level. The existing studies on these materials are generally associated with their synthesis, characterization for their complexes. There are very few methods of determination for the most widely used heavy metals such as lead and zinc. In this regard, the aim of this thesis work is the development of rapid and practical spectrophotometric methods of determination for lead. The analytical method for lead exploits the formation of its complex with 1,5-Bis (salisylidene)thiocarbohydrazone (BSTH) followed by its spectrophotometric determination at 419 nm. The analytical performance characteristics of the developed methods for the analytes as molar absorptivity (?); limit of detection (LOD) and limit of quantification (LOQ) are as follows: For Lead, ?=1,20x104 L mol-1cm-1; LOD=1,8x10-6 mol L-1 and LOQ=6x10-6 mol L-1; for Zinc, ?=2,54x104 Lmol-1cm-1; LOD=2,36x10-7 molL-1and LOQ= 7,88x10-7molL-1. In this study, the interfering effects of other metal ions, such as Ni(II), Cd(II), Zn(II), Ag(II), Cu(II), Hg(II) and Fe(III) ions in determination of Pb(II) were controlled using NaCN. Under the optimum conditions, the effects of various foreign ions on the determination of 10-3 M Pb(II) were examined seperately.
