İmmunohistokimya (IHK) ve Real-Time polymerase chain reaction (PCR) teknikleri EHV-1 hastalığının tanısında kullanılan önemli tanı yöntemleridir. Bu çalışmada aborte ölü ya da canlı doğup, doğumdan kısa bir süre sonra ölen taylara ait doku örneklerinde rutin histopatolojik teşhis, immunohistokimya (IHK) ve Real-Time PCR yöntemleri kullanılarak EHV-1'in varlığının araştırılması ve teşhis yöntemlerinin uyumluluğunun değerlendirilmesi amaçlandı. Bu amaçla İstanbul, İzmit, Bursa ve Eskişehir illerinden gönderilen 38 adet aborte fötuslara ait doku örnekleri (akciğer, karaciğer, dalak, böbrek, plasenta ve beyin) incelendi. Alınan doku örnekleri %10'luk formol saline solusyonunda tespit edilerek parafin bloklara gömüldü ve dokular gerekli laboratuar işlemlerinden geçirildikten sonra Rotary mikrotomuyla 3µm kalınlığında kesildi. Yine gerekli laboratuvar işlemlerinden geçirildikten sonra Hematoksilen-Eozin (H&E) ile boyandı ve ışık mikroskobunda incelendi. IHK için de dokular Avidin-Biodin peroksidaz metoduyla ilgili antikorlarla boyandı. Real-Time PCR yöntemi için -80°C'de dondurulmuş doku örnekleri kullanıldı. Sonuç olarak 38 adet taya ait doku örneklerinin Real-Time PCR ile %52,6'sının (20/38) pozitif; immumohistokimyasal yöntem ile %39,4'ünün (15/38) pozitif olduğu saptandı. İki tanı yöntemi ile elde edilen sonuçların çoğunlukla örtüştüğü gözlendi. Histopatolojik değerlendirmede ise 38 adet taya ait doku kesitinin %52,6'sinde (20/38), karaciğer çoğunlukta olmak üzere akciğer ve dalağa ait doku kesitlerinin en az bir tanesinde inklüzyon cisimciği saptandı. Immunohistokimya (IHK) ve Real-Time PCR yöntemleri ile pozitif olarak tespit edilen tayların dokularına ait lezyonların, hastalığın karakteristik bulgularının birçoğunu gösterdiği izlendi.Anahtar kelimeler: At, abort, EHV-1, immunohistokimya, Real-Time PCR.
Immunohistochemistry (IHC) and Real-Time Polymerase Chain Reaction (Real-Time PCR) are important ancillary diagnostic techniques in the detection of Equine Herpesvirus-1 (EHV-1) infections. The objective of the study was to investigate the incidence of EHV-1 infections in the tissue samples of aborted stillborn and live born foals and the foals which died soon after birth and compare and correlate the conventional histopathologic diagnosis with IHC and Real-Time PCR. The study material consisted of tissue samples (lungs, liver, spleen, kidney, placenta and the brain) belonging to 38 aborted foals submitted to our department from the vicinity of Istanbul, Izmit, Bursa and Eskisehir. The specimens were fixed in 10 % buffered formalin solution, routinely processed, cut at 3µm thickness and stained with Hematoxylin and Eosin Stain (H&E) and finally evaluated under light microscopy. Avidin-biotin peroxidase technique was applied for IHC. Samples were kept at -80°C?de for Real-Time PCR analysis. In conclusion Real-Time PCR revealed 52.6 % positivity (20/38), whereas 39.4 % (15/38) positivity was achieved with IHC, which indicated a correlation between these two diagnostic methods. Histopathology demonstrated 52,6 % (20/38) positivity in which tissue samples from at least one organ out of three (lungs, spleen and the liver) releaved inclusion bodies, the liver comprising the majority. Tissue samples of the foals found to be positive by IHC and PCR revealed histopathologic findings compatible with the characteristics of the disease.
