Sığır etlerinin, tüketicilerin istediği ölçüde kaliteli olmaması önemli bir sorundur. Bu sorun, çevre koşullarının yanı sıra hayvanın genotipi ile yakından ilişkilidir. Etin kalitesinin belirlenmesinde etkili olan en önemli özellikler etin gevrekliği ve etin mozaikleşmesidir. Et kalitesi üzerine çeşitli genetik çalışmalar yapılmış ve tek nükleotid polimorfizmi ile (SNP) doğrusal korelasyon ilişkileri saptanmıştır. Leptin (LEP) ile Tiroglobulin (TG) geni; mozaikleşme, sırt yağ kalınlığı, et tekstürü, et verimi ve et kalitesi ile ilişkilidir. Edirne'deki besi işletmelerinden 100 baş Türk Holstein sığır örneğinde LEP ile TG genlerinde çalışılmıştır. "Polimeraz Zincir Reaksiyonu–Restriksiyon Parça Uzunluk Polimorfizmi (PCR-RFLP)" metodu ile LEP geni 2. Ekzon da iki (E2JW, E2FB) ve TG geni 5' Promotor bölgesinde bir (TG5) SNP markörü incelenmiştir. Çalışmamızda genotiplendirme "Gelişmiş Analitik Fragman Analizörü Kılcal (Kapiller) Elektroforezi" metoduyla yapılmıştır. Et kalitesini belirlemede etkili olan bu üç markörle, mozaikleşme, renk analizi, pH ve etin gevrekliği ile ilgili SNP allelleri belirlenmiştir. Türk Holstein sığırlarında LEP E2JW'de 3 farklı genotip, LEP E2FB'de ise 2 farklı genotip gözlenmiştir. TG geni monomorfik bulunmuştur. Türk Holstein sığırının LEP ve TG geninde 3 SNP bakımından genetik karakterizasyonu belirlenmiştir. LEP E2JW'deki AA, AT ve TT genotipli sığırların frekansları sırasıyla 0,56, 0,38 ve 0,06 olarak gözlenmiştir. LEP E2FB'de CT ve TT genotipinde sığırlar gözlenmiştir ve frekansları sırasıyla 0,94 ve 0,06 olarak belirlenmiştir. TG5 (C422T) 'de CC genotipli sığırların frekansı 1 olarak bulunmuştur. LEP E2JW'deki A ve T allel gen frekansları sırasıyla 0,75 ve 0,25 olarak gözlenmiştir. LEP E2FB'de C ve T allel gen frekansları sırasıyla 0,47 ve 0,53 olarak belirlenmiştir. Türk Hostein ırkında LEP E2JW lokusunda pişmiş et rengi parlaklığı (L*) bakımından AA, AT ve TT genotipindeki sığır etlerinde istatistiksel olarak anlamlı bir ilişki tespit edilmiş olup farklar önemli bulunmuştur (p<0,05). Yedi gün boyunca bekletilip pişirilmiş etlerin parlaklıklığı, LEP E2JW lokusu AA ve AT genotipindeki sığır etlerin TT genotipindeki sığır etlerinden daha parlak ve farkın önemli olduğu tespit edilmiştir (p<0,05). LEP E2JW lokusundaki genotipik farklar bakımından Türk Holstein sığırları çiğ et b⃰ renk değeri önemli olup AA genotipindekiler AT ve TT genotipindekilere göre daha sarımtraktır (p<0,05). LEP E2JW lokusu 14. gün Musculus Longissimus Dorsi (MLD) pH ölçümleri bakımından genotipler arası fark anlamlı bulunmuştur (p<0,05). En yüksek pH değeri AT genotipinde ve en düşük pH değeri ise AA genotipinde gözlenmiştir. Yedinci günde pişmiş etlerin tekstürü yani kesilme kuvveti (SF) bakımından, LEP E2JW lokusu AA ve AT genotipindeki Türk Holstein sığırların MLD etlerinin TT genotipindekilerden farklı olduğu belirlenmiştir (p<0,05). Türk Hostein sığırlarında LEP E2FB lokusunda L* a* b* renk uzayı, SF ve pH bakımından CT ve TT genotipindeki sığır etlerinde istatistiksel olarak anlamlı bir ilişki tespit edilmemiştir (p>0,05). Türk Holstein sığırların CA, SKA ve MS ile LEP E2JW, E2FB ve TG5 markör genotipleri arasında olumlu bir ilişki kurulamamıştır (p>0,05). Yine TG TG5 lokusunda da L* a* b*, SF ve pH ile Türk Holstein sığırlarının genotipleri arasında olumlu bir ilişki kurulamamıştır. Sonuç olarak; Türk Holstein sığırlarında et gevrekliği için LEP E2JW lokusu T allelinin diğer araştırmacıların aksine olumlu etkisinin olmadığı anlaşılmış ve MDS çalışmalarına A allelinin olumlu katkısı olacağı söylenebilir. TG5/LEP E2JW/E2FB markör lokusları bakımından sırasıyla CC/AA/CT veya CC/AT/CT genotipindeki Türk Holstein sığırların daha kaliteli et ürettiği belirlenmiştir. It is an important problem that beef does not have enough quality that consumers request want. This problem is closely related to the animal's genotype as well as to environmental conditions. The most important characteristics that are effective in determining the quality of meat are the tenderness of meat and the mosaicization of the meat. Various genetic studies on meat quality were made and linear correlation correlations with single nucleotide polymorphisms (SNP) were determined. Leptin (LEP) and the thyroglobulin (TG) gene; is associated with mosaicization, back fat thickness, meat texture, meat yield and meat quality. LEP and TG genes of cattle were investigated in 100 head of Turkish Holstein in the beef feedlots of Edirne. Two SNP markers (E2JW and E2FB) in exon 2 of the leptin gene and one SNP marker (TG5) in 5' promoter region of TG gene were investigated with 'Restriction Fragment Length Polymorphism (PCR-RFLP)' method. In our study, genotyping was performed by "Advance Analytical Fragment Analyzer Capillary Electrophoresis" method. The SNP alleles with regard to marbling, colour analysis, pH and tenderness of the meat were determined with these three markers which are effective on detecting quality of the meat. Three different genotypes in LEP E2JW and 2 different genotypes in LEP E2FB were observed Turkish Holstein cattle. TG5 (C422T) was found monoforphic. Genetic characterization of 3 SNPs in LEP and TG gene of Turkish Holstein cattle were determined. The frequencies of AA, AT and TT genotyped cattle in LEP E2JW were observed as 0,56, 0,38 and 0,06, respectively. CT and TT genotypes were observed in LEP E2FB and their frequencies were determined as 0,94 and 0,06, respectively. The frequencies of CC genotyped cattle were found in TG5 (C422T) as 1. The frequencies of A and T alleles in LEP E2JW were observed as 0,75 ve 0,25, respectively. The frequencies of C and T alleles in LEP E2FB were determined as 0,47 and 0,53, respectively. In the LEP E2JW loci of the Turkish Holstein breed, with regard to cooked meat color brightness(L*) statistically significant correlation was found in the beef with AA, AT and TT genotype (p<0,05). On account of the brightness of cooked meat after waiting during seven days, LEP E2JW locus with AA and AT genotypes of beef is brighter than TT genotype and it was determined that the difference is significant (p<0,05). In point of the genotypic differences in LEP E2JW loci, uncooked meat b⃰ color value of Turkish Holstein cattle is important (p<0,05) and with AA genotypes are more yellowish than with AT and TT genotypes. The difference between genotypes was found significant with regard to the fourteenth day pH measurements of Musculus Longissimus Dorsi (MLD) with LEP E2JW loci. The highest pH value was observed in AT genotype and the lowest pH was observed in AA genotype. In LEP E2JW loci, texture of cooked meats on seventh day that is with regard to shear force (SF) value, MLD meats of Turkish Holstein cattle with AA and AT genotype were found to be different from TT genotype (p<0,05). In LEP E2FB loci of theTurkish Hostein cattle, L*a*b* color space, SF and pH in CT and TT genotype was not found to be a statistically significant relationship (p>0,05). There was no positive relationship between CA, SKA and MS of Turkish Holstein cattle and LEP E2JW, E2FB and TG5 marker genotypes (p>0,05). Again, in the TG TG5 loci there was no positive relationship between L*a*b*, SF and pH and genotypes of Turkish Holstein cattle. Concequently; It was understood that the T allele of the LEP E2JW loci for meat texture in Turkish Holstein cattle has no positive effect as opposed to other researchers and it can be said that the A allele will contribute to the MDS studies. It has been identified that Turkish Holstein cattles with the CC/AA/CT or CC/AT/CT genotypes produce higher quality meat for TG5 / LEP E2JW / E2FB marker loci.
